From what I see, in the Screenshot shortcut settings, I could only choose - Full screenshot, Area screenshot, and copy screenshot to the clipboard. I'm only interested in area screenshot but it's also tedious to copy and paste manually each screenshot.
I tried to use a custom shortcut but I can't see any option for save, ("io.elementary.screenshot-tool -r -c" does most what I want but saving, and vice versa "io.elementary.screenshot-tool -r" does most what I want but copying).
It is possible to add an additional option - "-v" (for save)?

I'm confused. You can use any of the three modes (screen, app, or area) with either of the save options (copy or save to disk). Are you asking for the ability to copy and save to disk, or something else?
